細胞載體於生物反應器中的使用已是耳熟能詳之商品,舉例像是fibra-cel® 、cytodex 等,雖然這些商品穩定性高,但價格卻過於昂貴且大多是進口,造成國內生技業者一直無法降低產品價格,也阻礙了國內生技產品,特別是生物製劑,無法打入國際市場與大陸市場,故開發一低成本載體是目前迫切需求的趨勢。聚氨酯(Polyurethane ; PU)載體,稱PU載體具備低密度、貼附之表面積大、易吸收水分且其成本價格低廉之優點,成為本實驗室所研究之新型載體。在此研究中,藉由非洲綠猴腎細胞((African green monkey kidney cell line ; Vero)生產腸病毒71型 (EV71) 病毒為例,利用低成本之PU載體分別以攪拌式及潮汐式量產方式培養細胞,以生產腸病毒 EV71。透過細胞載體的改良,希望能運用於疫苗生產程序以能降低成本,符合市場需求。

The use of carrier for animal cell cultivation in the bioreactor is already familiar, such as fibra-cel®, cytodex etc. Although these cell carriers are quite stable in its use, the high prices cause the tendency to develop a new carrier. Polyurethane (PU) its properties of low density, large surface area for attachment, ease to absorb moisture and low cost has become advantage to be developed as a new cell carrier. In this study, african green monkey kidney cells used as an example was cultivated in PU carriers in stir-tank reactor and tide bioreactor for Enterovirus 71 production. The productions of cell and virus were estimated. It is expected that the developed new carriers can be applied to vaccine production process to reduce the vaccine cost, and match the market needs.
